Further studies on microbiological ring-expansion of penicillin N.
The rate of microbiological ring-expansion of penicillin N to deacetoxycephalosporin C using protoplast lysates of the antibiotic-negative mutant Cephalosporium acremonium M-0198 has been increased some 70-fold over that of our earlier system. We confirmed the stimulatory effects of FeSO4 and ascorbate described by Hook et al. (Biochem. Biophys. Res. Commun. 87: 258, 1979); the optimum concentr...
متن کاملOf Antibiotics Microbiological Ring Expansion of Penicillin
A mutant of Cephalosporium acremonium producing high amounts of penicillin N was isolated. This antibiotic was purified and characterized. It was possible to convert this penicillin N to deacetoxycephalosporin C enzymatically. The reaction could be carried out with enzyme systems prepared from C. acremonium mutants producing either no S-lactam antibiotics or excreting only penicillin N. It was ...
متن کاملCell-free ring expansion of penicillin N to deacetoxycephalosporin C by Cephalosporium acremonium CW-19 and its mutants.
To examine microbiological ring expansion of penicillin N to a cephalosporin, we obtained five mutants of Cephalosporium acremonium blocked in beta-lactam antibiotic biosynthesis from 2500 survivors of mutagenesis. In submerged fermentation, mutants M-0198, M-0199, and M-2351 produced no beta-lactam antibiotic (type A), whereas mutants M-1443 and M-1836 formed penicillin N but not cephalosporin...
متن کاملStability and hyperstability of orthogonally ring $*$-$n$-derivations and orthogonally ring $*$-$n$-homomorphisms on $C^*$-algebras
In this paper, we investigate the generalized Hyers-Ulam-Rassias and the Isac and Rassias-type stability of the conditional of orthogonally ring $*$-$n$-derivation and orthogonally ring $*$-$n$-homomorphism on $C^*$-algebras. As a consequence of this, we prove the hyperstability of orthogonally ring $*$-$n$-derivation and orthogonally ring $*$-$n$-homomorphism on $C^*$-algebras.
متن کاملEngineering Streptomyces clavuligerus deacetoxycephalosporin C synthase for optimal ring expansion activity toward penicillin G.
The deacetoxycephalosporin C synthase (DAOCS) from Streptomyces clavuligerus was engineered with the aim of enhancing the conversion of penicillin G into phenylacetyl-7-aminodeacetoxycephalosporanic acid, a precursor of 7-aminodeacetoxycephalosporanic acid, for industrial application. A single round of random mutagenesis followed by the screening of 5,500 clones identified three mutants, G79E, ...
